Supplemental Figure 1 -
Supplemental Figure 1. GST-CCA1 binds with similar affinity to EEmt and CBS sequences
(A) Sequences used to compete for binding. (B) GST or GST-CCA1 containing extracts were incubated with radiolabeled double stranded DNA containing a CCR2_EE site. 5, 15, 50, 150, or 500-fold molar excess of unlabeled competitor DNA was added to each reaction as indicated. DNA and protein/DNA complexes were separated by nondenaturing gel electrophoresis and visualized using a phosphorimager. The arrowheads indicate the position of unbound probe.
Supplemental Figure 2 -
Supplemental Figure 2. GST-LHY has similar binding preferences as recombinant CCA1
(A) Locations of bases that were altered to make each indicated mutant. Actual sequences of competitors used can be found in Figure 5. (B) Extracts expressing GST or GST-LHY were incubated with radiolabeled double stranded DNA containing a CCR2_EE site. 5, 15, 50, 150, or 500 molar excess of unlabeled competitor DNA was added to each reaction as indicated. Protein/DNA complexes were separated by nondenaturing gel electrophoresis and visualized using a phosphorimager. The arrowheads indicate the position of unbound probe.
